Kishan Dholakia will talk on the topic “Structured Light Sheet Microscopy: advances in application and analysis”.
Abstract:
We have seen great advances in photonics-based methods for imaging. However, whilst improvements have been seen in beating the diffraction limit, obtaining wide field imaging and imaging through scattering media remain a challenge. I will describe approaches that tackle this issue for light sheet imaging or selective plane illumination microscopy.
For the goal of a wider field of view, I will describe the use of shaped fields namely propagation invariant (‘non-diffracting’) light fields and complex beam shaping for light sheet imaging. Propagation invariant light fields retain their transverse intensity profile upon propagation. More recent advances using attenuation compensation of fields [1], three photon light sheet imaging [2] and new sample mounting procedures [3] using acoustic confinement will be highlighted.
More recently we have explored the use of deep learning. We have developed an unsupervised approach that is trained on a single light sheet volume dramatically reducing the image data needed [4]. This can be combined with shaped light for enhanced imaging performance.
